Treatment protocol Mice were infected with LCMV Arm and LCMV CL13 by intraperitoneal and intravenous injection, respectively.
Extracted molecule total RNA
Extraction protocol Splenic CD4+CD25+ regulatory T (Treg) cells were isolated from naïve and infected mice at 0 and 16 day after acute and chronic virus infection using a CD4+CD25+ regulatory T Cell Isolation Kit (MiltenyiBiotec). Total RNA was extracted from isolated Treg cells using the RNeasyPlus Micro Kit (Qiagen). RNA purity was confirmed through the Agilent Bioanalyzer 2100 and nano drop 8000 analysis.
Label Biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 300 ng total RNA (GeneChip® 3’IVT Express kit P/N 901229, Affymetrix).
 
Hybridization protocol Following fragmentation, fragmented cRNA were hybridized for 16 hr at 45C on AffymetrixGeneChip® HT MG-430 PM Array plate. The GeneChips were washed and stained in the Affymetrix GeneTitan® MC Instrument.
Scan protocol GeneChips were scanned using the Affymetrix GeneChip® HT Scanner.
Description Biological replicate 2 of 3, CD4+CD25+ Treg cells, 16 d after chronic infection, isolated from spleen using CD4+CD25+ regulatory T Cell Isolation Kit (MiltenyiBiotec)
Data processing The Micorarray Suite version 5 (MAS5) algorithm implemented in Affymetrix Expression Console software (version1.3.1) was used to normalize the raw data. Highly expressed genes that showed over 2-fold difference between the signal values of control and test samples were selected for further study.
