Treatment protocol |
Epididymal adipose tissue was washed by cold RNase-free normal saline, quickly dissected and put in liquid nitrogen when sacrificing the mice. Tissues were stored in -80°C until RNA extraction.

All samples were prepared using Trizol. Total RNA was isolated using the RNeasy Mini Kit (QIAGEN), cDNA synthesis and amplification was performed using the WT cDNA synthesis and amplification kit (Affymetrix).

Biotinylated cDNA were prepared according to the standard Affymetrix protocol from 250 ng total RNA by using Ambion® WT Expression Kit.

Hybridization protocol |
Following labeling, 5.5 ug of cDNA were hybridized for 16 hr at 45°C on GeneChip Mouse Exon 1.0 in Hybridization Oven 645. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

Data processing |
The data were analyzed with Robust Multichip Average (RMA) using Affymetrix Expression Console default analysis settings and global scaling as normalization method. Normalized signal intensities were transformed to the log base 2 scale.
