Treatment protocol |
testes without seminal vesicle and accessory glands were dissected in PBS for 30 minutes and immediately frozen in liquid nitrogen before transferred to -80°C. Total RNA was extracted using TRIzol (Invitrogen). RNA quality was assessed by checking rRNA integrity using Bioanalyzer. ~250ngs of total RNA was processed for each replicate.

Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Drosophila Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

GeneChips were scanned using the Affymetrix Scanner 3000-7G
|
Data processing |
Data were background adjusted and normalized using R program (v3.0.2) with the BioConductor package GCRMA.
