Double stranded RNA oligonucleotides are transfected to cells with lipofectamine 3000 (Invitrogen) according to the manufacturer’s protocols. RNA was extracted 48hours after transfection.

DICER exon 5-disrupted RKO cells are were maintained in Roswell Park Memorial Institute (RPMI) 1640 Medium supplemented with 10% fetal bovine serum (FBS) (Thermo Fisher Scientific) and grown at 37°C in a humidified incubator with 5% CO2.
