Growth protocol |
Cells were grown in minimal medium at 37°C to mid exponential phase. Cells were treated with a formaldehyde-containing fixing solution, washed and lysed in the presence of protease inhibitors.

Lysed cells were sonicated and cleared lysates were kept as the input samples. The IP samples were incubated with Protein G coupled dynabeads (Invitrogen), coupled to the respective antibodies (Smc, ParB or ScpB). DNA was purified by phenol chloroform extraction and ethanol precipitation.
