Treatment protocol Melanosomes of WM3682 and WM3314 cells were treated with RNase for 10 min at 37 °C, followed by the addition of RNase inhibitor.
Growth protocol MNT-1, WM3682 and WM3314 cells were cultured in DMEM medium supplemented with 10% FBS, 1% penicillin-streptomycin, and 1% L-glutamine.
Extracted molecule total RNA
Extraction protocol RNA of WM3682 and WM3314 cells and melanosomes was extracted with Trizol reagent. RNA from MNT-1 exosomes and secreted melanosomes was isolated using the miRNeasy Mini Kit from Qiagen. Quality control was performed with Agilent Bioanalyzer.
Label Cy3
Label protocol Samples were labeled according to the Agilent miRNA Complete Labeling and Hyb kit protocol.
 
Hybridization protocol Hybridization was performed in a SureHyb chamber at 55°C for 20h.
Scan protocol Microarray scanning was performed using the Agilent Scanner G2505C.
