Title |
Melanosomes WM3314_rep1 |
Sample type |
RNA |
Source name |
WM3314 melanosomes
Organism |
Homo sapiens |
Characteristics |
cell line: WM3314 cell component: melanosomes
Treatment protocol |
Melanosomes of WM3682 and WM3314 cells were treated with RNase for 10 min at 37 °C, followed by the addition of RNase inhibitor.
Growth protocol |
MNT-1, WM3682 and WM3314 cells were cultured in DMEM medium supplemented with 10% FBS, 1% penicillin-streptomycin, and 1% L-glutamine.
Extracted molecule |
total RNA |
Extraction protocol |
RNA of WM3682 and WM3314 cells and melanosomes was extracted with Trizol reagent. RNA from MNT-1 exosomes and secreted melanosomes was isolated using the miRNeasy Mini Kit from Qiagen. Quality control was performed with Agilent Bioanalyzer.
Label |
Cy3
Label protocol |
Samples were labeled according to the Agilent miRNA Complete Labeling and Hyb kit protocol.
Hybridization protocol |
Hybridization was performed in a SureHyb chamber at 55°C for 20h.
Scan protocol |
Microarray scanning was performed using the Agilent Scanner G2505C.
Description |
RNA from melanosomes of WM3314 cells MEL 3314
Data processing |
The data were quantile normalized and fold-changes were calculated using R/Bioconductor scripts.
