Characteristics UET-13TR-EWS/ERG cells were cultured in the presence of tetracycline for 24 hours
Treatment protocol UET-13 transfectants were treated with or without 3 μg/ml of tetracycline for the indicated periods.
Growth protocol UET-13 transfectants (UET-13TR-EWS/FLI1 and UET-13TR-EWS/ERG ) were cultured in Dulbecco’s modified Eagle’s medium (DMEM) with 10% Tet System Approved Fetal Bovine Serum (T-FBS)(Takara, Shiga, Japan) at 37°C under a humidified 5% CO2 atmosphere.
Extracted molecule total RNA
Extraction protocol Total RNA was extracted from cells using RNeasy kit (QIAGEN, Valencia, CA) according to the manufacturer's instructions.
Label Biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 5 ug total RNA.
 
Hybridization protocol Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on Affymetrix HG-U133 Plus 2.0. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol GeneChips were scanned using the GeneArray Scanner GCS3000.
Description Gene expression data from UET-13TR-EWS/ERG cells incubated in the presence of tetracycline for 24 hours
Data processing The arrays were analyzed using GeneChip Operating Software 1.2 (GCOS 1.2) (Affymetrix). Background subtraction and normalization were performed by GeneSpring GX 7.3 software (Agilent Technologies, Inc., Santa Clara, CA).
