Sorted cells were lysed with TRIzol and stored at -80 degree. RNA was extracted according to the manufacturer’s manual (Life Technologies). cDNA libraries were generated using SMARTer amplification (Clontech) and individually indexed. Libraries were sequenced on HiSeq 2000 platform (Illumina).

Description |
LCMV-specific GP66-tetramer+ memory CD4 T cells were isolated 60 days p.i and transferred to congenically marked recipients. Recipients were infected with LCMV and 60 days later primary and secondary memory T cells were isolated. Naïve CD4 T cells from recipient mice were used as controls
