Treatment protocol |
0.5 nmol miR-132 mimics or control mimics (ThermoFisher Scientific) were mixed with MaxSuppressor™ In Vivo RNA-LANCEr II (Bioo Scientific) in a total volume of 100 μl for each wound and injected intradermally into 4 points around the wound edges in the mice immediately after wounding. Following the surgical procedure, the animals were individually caged. During the first 2 days, the animals received s.c. buprenorphine (0.03 mg/kg) twice a day for relief of any possible distress caused by the procedure.

Growth protocol |
Eleven-week-old male diabetic db/db [BKS(D)-Leprdb/JOrlRj)] mice on the C57Bl/6J background (Janvier Labs) were caged individually for one week, handled daily, and then wounded. General anesthesia was performed with 3% isoflurane (Abbott). The hair on the back was shaved with an electronic hair remover followed by Veet® hair removal cream. The skin was rinsed with PBS and two full-thickness wounds extending through the panniculus carnosus were made on the dorsum on each side of midline, using a 4 mm biopsy punch.

The mice were sacrificed 6 days after injury and the total RNA was extracted from wound-edge biopsies using miRNeasy Mini kit (Qiagen) according to the manufacturer's instructions.

150ng of total RNA were used to prepare cDNA following the GeneChip WT PLUS Reagent Kit labeling protocol.

The samples were hybridized to a mouse Clariom™ D assay plate (previously known as GeneChip® Mouse Transcriptome Assay 1.0) according to standard protocol.
