Treatment protocol |
Control and Vis-KO mice were housed at room temperature (22°C) until 6 weeks of age then transferred to thermoneutral housing conditions (30°C) for 2 weeks. After 2 weeks at thermoneutrality, mice were treated with PBS (vehicle) or CL316,243 (1 mg/kg, intraperitoneal) daily for 3 days. On the 4th day, tissues were harvested for analysis.

Growth protocol |
Mice were maintained on a 12 hr light/dark cycle in a temperature-controlled environment (room temperature, 22°C; thermoneutrality, 30°C). Mice were given free access to food and water, and maintained on a standard chow diet.

Extraction protocol |
RNA was extracted with Trizol and purified through ethanol precipitation. The library was constructed by the UT Southwestern McDermott Sequencing Core using the Illumina TruSeq Stranded mRNA Sample Preparation protocol.

Data processing |
RNA-sequencing was performed by the UT Southwestern McDermott Sequencing Core on an Illumina HiSeq 2500 v3 Sequencing System. Data was processed by the UT Southwestern McDermott Center Bioinformatics Core. Stranded, single-end 50-base-pair reads were generated. Reads were aligned using TopHat2 and Bowtie2. Raw counts were generated using Cufflinks and featureCounts. Cuffdiff and edgeR were used for differential expression analysis.
