Treatment protocol Cells were seeded into tissue culture plates such that cells would be 80-90% confluent by the end of the experiment. Cells were allowed to attach overnight in OSE media. The next day, cells were washed with PBS and the media was changed to hormone-free media consisting of 5% charcoal stripped FBS in phenol red free DMEM-F12, 2ng/mL epidermal growth factor, and 0.01mg/mL insulin-transferrin-sodium selenite supplement. Cells were allowed to normalize to hormone-free media for 48 h before E2 treatment. Cells were treated with 100nM E2 for 24H. An equivalent volume of 100% EtOH (vehicle) was added to control plates for a final concentration of 0.0002% EtOH.
Growth protocol MASE cells were maintained at 37°C in OSE media that consists of α-MEM media with 5% fetal bovine serum, 2ng/mL epidermal growth factor, and 0.01mg/mL insulin-transferrin-sodium selenite supplement.
Extracted molecule total RNA
Extraction protocol microRNA was isolated from cells using a miRNeasy miRNA isolation kit from Qiagen.
Label Biotin
Label protocol microRNA was labelled with the FlashTag Biotin HSR RNA labeling kit from Affymetrix.
 
Hybridization protocol Samples were prepared for Affymetrix GeneChip miRNA 4.0 Array according to the GeneChipTM miRNA Array Procedure (P/N 703095).
Scan protocol GeneChip miRNA 4.0 Arrays were scanned with the Affymetrix GeneChipTM Scanner 3000.
Data processing Affymetrix Transcriptome Analysis Console Software 3.0 was used to analyze the resulting array data.
