Treatment protocol |
Hair follicles obtained by FUE technique(0.8mm, 0.9mm, 1mm wide) were immediately placed in a sterile petri dish containing normal saline at 4℃.With an operating microscope and microsurgical instruments, fat and redundant tissues were removed from the hair follicle gently.Then two parts were cut away from the tissue: end-bulb areaand bulge region(4-6 mm below thesebaceous gland). The separated tissues were collected in TRIZOL Reagent(Invitrogen, Carlsbad, CA, USA) for next test.

Extraction protocol |
Total RNA was isolated using Trizol reagent (Invitrogen, Carlsbad, CA, USA) following manufacturer’s recommendations. RNA quality and quantity of all samples were evaluated by using Bioanalyzer 2100 and RNA 6000 Nano LabChip Kit (Agilent, CA, USA) with RIN number > 7.5. RNA libraries were prepared for sequencing using standard Illumina protocols

Illumina bcl2fastq software used for basecalling. Sequenced reads were trimmed for adaptor sequence, and masked for low-complexity or low-quality sequence, then mapped hg38 whole genome using tophat2 with parameters -read-mismatches 2 -read-edit-dist 2 -max-intron-length 5000000 -library-type fr-unstranded -num-threads 8 -mate-inner-dist 40. Fragments Per Kilobase of transcript per Million fragments mapped (FPKM) were calculated using a protocol from Trapnell et al., Nature Biotechnology, 2010.
