For the establishment of transgenic populations, media was supplemented with Puromycin (Sigma, 80µg/mL) or G418 (Gibco, 100µg/mL). Cultures of 80% confluent cultures were harvested between passages 2 and 4 for microarray analysis.

Growth protocol |
Control, SOX7 O/E and SOX17 O/E HESC were cultured under standard tissue culture conditions on mitotically inactivated mouse embryo fibroblast feeder layers in Knockout DMEM (Gibco) supplemented with 15% Serum Replacement (Gibco), Glutamax (Invitrogen), penicillin/streptomycin (Gibco), 1 mM non-essential amino acids (Gibco), 0.5mM mercaptoethanol, and 10ug/mL recombinant FGF2 (Peprotech).
