One day prior to the experiment (age: 8 days), velcro rings were glued to the feathers around the eye under diethylether anaesthesia. At the age of 9 days, after 4-6 hours of light-onset, plus 6.9 diopter lenses were fitted to both eyes of the plus_lens treated group, control chicks received no lenses. 24 hours later, Chicks were killed by an overdose of diethylether, decapitated and the eyes were enucleated. Retinal samples were snap-frozen in liquid nitrogen and stored at -80°C.

Growth protocol |
chicks were raised in groups under a 12-hr light/12-hr dark cycle with free access to food and water. Illumination was provided by light bulbs that produced an ambient illuminance of about 1000 lux on the cage floor.

The retinae of both eyes were pooled and RNA was extracted using the RNeasy Mini Kit (Qiagen, Hilden, Germany), according to the manufacturer’s instruction. RNA was quantified and checked for quality with the Agilent 2100 Bioanalyzer (Agilent Technologies, Palo Alto, CA). The RNA integrity numbers (RIN) ranged from 9.2 to 9.5 (with 1 being the most degraded profile and 10 being the most intact).

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 1.5 ug total RNA (see manufacturer´s website: Expression Analysis Technical Manual, One-Cycle Target Labeling Assay, Affymetrix).
