Extraction protocol |
CAMs were immediately snap-frozen in liquid nitrogen after excision. Total RNA was isolated using RNeasy mini kits (Qiagen) according to the manufacturer's instructions and quantified using a NanoDrop spectrophotometer.

Complementary RNA (cRNA) was prepared according to the manufacturer’s (Affymetrix) 'standard protocol'.

Following fragmentation, cRNA was hybridized for 16 hr at 45C on the GeneChip Chicken Genome array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

Description |
We investigated vascular development and maturation in vivo by determining global gene regulation throughout the formation of the chick chorio-allantoic membrane (CAM).

Data processing |
Data were analyzed with the GCOS 1.2 software (Affymetrix), using the default analysis settings and global scaling as normalization method, with a trimmed mean target intensity value (TGT) of each array arbitrarily set to 100.
