Characteristics |
Only sedentary male subjects were recruited. Patients with type 2 diabetes were either treated by diet alone or in combination with sulphonylurea or metformin, which were withdrawn one week prior to the study. The patients were all GAD65 antibody negative and without signs of diabetic retinopathy, nephropathy, neuropathy or macrovascular complications.

10 µg of fragmented cRNA were hybridised to a U95Av2 GeneChip (Affymetrix) using their standard procedure (45°C, 16-h). Washing and staining was performed in a Fluidics Station 400 (Affymetrix) using the protocol EukGE-WS2v4.

Data processing |
Microarray normalisation and calculation of expression measures were performed using Robust Multiarray Average, implemented in the statistical package R provided by Bioconductor (Seattle, WA, USA). The expression values were log 2 transformed to obtain a normal distribution across arrays and samples. Before analysing the expression data we removed Affymetrix control probes, resulting in a total of 12,558 probes.
