Extraction protocol |
DNA was extracted using a phenol/chloroform/isoamyl alcohol extraction after proteinase K treatment. DNAs were digested with HindIII and ligated to appropriate adapters prior to PCR amplification. Where DNA quantity was limited, a whole genome amplification step (Multiple Displacement Amplification) was performed prior to digestion according to manufacturer’s instructions (Quiagen Inc., Valencia, CA).
