st15 embryos: 14-16h embryos were inspected under the stereoscope and were selected for the desired stages and for Sxl-GFP presence or absence to distinguish their gender. 25 embryos were homogenized in Trizol and total mRNA was isolated using the microRNA Zymo Kit. RNA Quality was assessed by Fragment Analyzer. TruSeq Stranded mRNA.
