Treatment protocol |
BJ-hTERT cells were immortalized by stable transfection with pBabe-hTERT. Replicative senescence was confirmed by morphology change, beta-gal stainng, and growth curve plateau.

Fibroblasts were cultured at 37 degrees and 5% CO2 in MEMĪ± supplemented with 1mM sodium pyruvate, 1.5 g/L sodium bicarbonate, and 10% fetal bovine serum.

miRNA expression in early passage immortalized fibroblasts

Data processing |
Mean signal for each probe was quantile normalized and the log2 was taken. Probes were collapsed to individual miRNAs by averaging. Each cell line high passage miRNA array was normalized to its corresponding low passage miRNA array.
