Total RNA was extracted from skin biopsies by the Qiagen miRNeasy Mini Kit (Valencia, CA, USA). DNA was removed with on-column DNase digestion by the Qiagen RNase-free DNase Set. The quality of extracted RNA was examined using Agilent Bioanalyzer 2100 (Agilent Technologies, Palo Alto, CA).
Libraries were prepared using TruSeq Stranded mRNA Library Prep kit (Illumina, San Diego, CA), according to the manufacture’s instructions. The normalized cDNA libraries were pooled and sequenced on Illumina Novaseq 6000 with pair-end 100 cycles in 50M reads sequencing depth. Image analysis and bases calling were conducted in real-time by the Illumina analysis pipeline.
