Extraction protocol |
Somatosensory cortex was microdissected from 400 micron thick slice. The microdissected tissue chunks were dissociated by pronase E and trituration. Fluorescently labeled neurons from the dissociated cells were hand-sorted under a fluorescent dissection microscope using glass pipette with 50 micron diameter and lysed in XB buffer (Arcturus). Total RNA was extracted from the lysate using PicoPure Kit (Arcturus). (Detailed protocol is available at http://mouse.bio.brandeis.edu/protocol.pdf .)
