Extraction protocol |
Spleen and small intestine were processed to single cell suspensions. Cells were antibody stained for Flourescence activated cell sorted. Viability dye and antibodies against CD4, CD8, TCRb, and B220 were utilized to sort live TCRb+CD4+CD8-B220-GFP+ Treg cells and TCRb+CD4+CD8-B220-GFP- T conventional cells. RNA was extracted with Trizol reagent, followed by clean-up and with QIAGEN RNeasy mini kit before labeling and microarray preparations. Each sample includes cells from multiple mice to reach sufficient intestinal Treg numbers for RNA isolation
