CsA (Sigma) was added to some cultures at 2 µg/ml for 30 min prior to addition of Flt3-L and was maintained throughout the culture.

Growth protocol |
Bone marrow cells harvested from femurs and tibiae of 8-12 week old C57BL/6 mice were cultured in Iscove’s modified Dulbecco’s medium (containing 10% heat-inactivated FCS (Life Technologies), penicillin 100U/ml, streptomycin 100 ug/ml and Flt3-L ~200 ng/ml (from transfected B16 tumor cells (51)) at 2.5x 106 cells/ml.

Microarray data handling was mainly done using Bioconductor 2.6 and an R package AMDA. The Robust Multichip Average (RMA) method was employed to calculate probe set intensity.
