Extraction protocol After harvest and cleaning, ganglionic tissue was incubated in artificial cerebrospinal fluid (ACSF) oxygenated by Carbogen with a stabile pH of 7.4 at 37°C for 2 hours. SCGs were then dissociated to single cells with collagenase II and proceeded for single cell capture, barcoding and library preparation (10x genomics).
Barcoding and library preparation was done using Chromium single cell 3’ (v3, 10x Genomics).
