Extracted molecule total RNA
Extraction protocol Total RNA was extracted from myeloid dendritic cells with TriZol reagent (Invitrogen) according to a standard protocol. RNA integrity of individual samples was confirmed with an Agilent 2100 Bioanalyzer. CD11c+ cells were purified to 97% purify from lung mononuclear cells by two consecutive rounds of magnetic cell separation using CD11c-antibody-labled magnetic beads and LS columns for positive cell selection as per manufacturer’s protocol (Miltenyi Biotech). The purity of CD11c+ cells was verified by flow cytometry prior to pooling CD11c+ cells from three mice for total RNA isolation. Two pools of CD11c+ cells from three mice each were prepared for WT and Il1a-/- experimental groups.

Label protocol For microarray hybridization, RNA was processed for hybridization to Affymetrix Mouse Gene 1.0 ST Array through a core facility in the Functional Genomics Laboratory in the Center for Ecogenetics and Environmental Health at the University of Washington according to the manufacturer's instructions.
 
Hybridization protocol The standard hybridization protocol was used as recommended by Affymterix.
Scan protocol GeneChips were scanned using the Affymetrix GeneArray Scanner 3000 through the University of Washington Functional Genomics Laboratory in the Center for Ecogenetics and Environmental Health (http://depts.washington.edu/ceeh/ServiceCores/FC1/FC1.html).
