Title |
TAF7 Depletion in Mouse Embryonic Fibroblasts, sample 107 |
Sample type |
RNA |
Source name |
embryonic fibroblasts, Taf7(+/+)-2, infected, 48 hrs
Organism |
Mus musculus |
Characteristics |
background strain: C57/BL6 and 129 mix cell type: mouse embryonic fibroblast (MEF) genotype: Taf7(+/+) mscv cre-gfp retroviral infection: yes time recovery (hour): 48
Growth protocol |
The MEFs are grown in DMEM with 10% Fetal Calf Serum, Hepes 20mM, and gentamycin.
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA is extracted using shredders and the Qiagen RNeasy mini kit. Their quality was assayed before using for hybridization.
Label |
biotin
Label protocol |
Labeling was performed according to the Affymetrix (Santa Clara, CA) GeneChip Whole Transcript (WT) Sense Target Labelling Assay manual.
Hybridization protocol |
Microarrays were hybridized overnight with 5 ug biotin-labelled ss-DNA, and washed in fluidics station wash protocol: FS450_0001.
Scan protocol |
GeneChip scanner 3000 7G was used.
Description |
m107 Taf7(+/+)-1 and Taf7(+/+)-2 represent 2 different wild-type MEF cell lines.
Data processing |
Exon array data were analyzed with Affymetrix Expression Console SoftwareTM (version 1.1). The Robust Multi-array Analysis (RMA) algorithm was used for gene intensity analysis. Only genes in the “core” set, which represents RefSeq and full-length GenBank mRNAs, were included in the analysis.
