Treatment protocol |
Brains and imaginal discs were dissected in PBS under a dissecting scope and tissues from 10 larvae per sample were collected in a tube on dry ice.

Growth protocol |
Flies were kept on standard fly food at 25 degrees. Third instar wandering larvae were used for dissections.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 500 ng total RNA using the Affymetrix 3’ IVT Express kit.

Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridised on Affymetrix Drosophila 2.0 GeneChips for 16 hrs at 45°C. Gene chips were washed and stained with streptavidin-phycoerythrin using the Affymetrix Fluidics Station 450.

Scan protocol |
GeneChips were scanned using an Affymetrix GeneChip 3000 scanner.

Gene expression data from mitotic Drosophila cells that have too many centrosomes

Data processing |
The data was analysed using the Bioconductor package limma. Probe intensities from Affymetrix image files (.CEL files) were normalized using quantile normalisation, and expression signals of all probesets were calculated using GCRMA.
