Design: Library construction 1) Collect and purify genomic DNA; 2) Genomic DNA fragmentation using Covaris to construct genome sequencing libraries; 3) Connect A & B connectors; 4) Screening and removal of adapter self-connecting fragments; 5) Agarose gel electrophoresis was used to screen the fragments, and the fragments with one end of the A adapter and the other end of the B adapter were retained; 6) Sodium hydroxide is denatured, resulting in single-stranded DNA fragments.
