SRX8979431: Metatranscriptome of Wasmannia auropunctata: Adult worker

Design: Total RNA was obtained from Wasmannia auropunctata ants by the Trizol and PureLink methods. mRNA from was enriched using oligo(dT) beads and rRNA removed using the Ribo-Zero kit. Library construction was completed by randomly fragmenting the mRNA. cDNA was synthesized using mRNA template and random hexamers primers, after which a custom second-strand synthesis buffer (Illumina), dNTPs, RNase H and DNA polymerase I were added to initiate the second-strand synthesis. Sequence adapters were ligated to the cDNA, the double-stranded cDNA library was completed by size selection and PCR enrichment. The qualified libraries were sequenced on an Illumina (Miseq chemistry) sequencer after pooling according to its effective concentration and expected data volume.
