SRX19145819: DNA-Seq of Rouxiella badensis: DSM 100043
1 OXFORD_NANOPORE (GridION) run: 426,035 spots, 2.3G bases, 1.9Gb downloads

Design: A Single colony was grown overnight in PD broth at 28C with shaking at 200 rpm. The culture was then spun down and the pellet was used for DNA extraction through Promega DNA extraction kit. After checking the quality of the DNA it was used for long read Oxford Nanopore. The long read sequencing was generated by Garvan Institute using an Oxford Nanopore GridION platform with FLO-MIN106 flow cell. The SQK-LSK109 kit was used without shearing and the reads were called and quality controlled during sequencing using MinKNOW version 20.06.17 and Guppy version 4.0.11. This generated total of 268,783 reads.

In this research we announce the complete assembly of a new Australian strain of the Gram-negative bacterium R. badensis DAR84756 and the complete assembly of the German type-strains, R. badensis DSM100043T and R. chamberiensis DSM28324T. The genomes were resolved using ONT long DNA reads and Flye assembler with a mean coverage of over 400x for each genome.
