Design: Parasites were liberated from host cells by saponin lysis. Genomic DNA was extracted by cell lysis with SDS/proteinase K, followed by phenol:chloroform extraction and ethanol precipitation as previously described. Genomic DNA pellets were resuspended in 10 mM Tris, 1 mM EDTA and assessed by agarose gel electrophoresis and Nanodrop spectrometry. The purified genomic DNA samples were shipped on dry ice to NovogeneAIT (Singapore) for whole-genome sequencing service.

Study: 9-methoxystrobilurin-G induced resistance

The natural product 9-methoxystrobilurin-G (9MG) from Favolaschia spp basidiomycetes is a potent and selective antimalarial. The mechanism of action of 9MG is unknown, but structurally related strobilurins from other fungal species are known to inhibit mitochondrial cytochrome b (CYTB) complex III. We induced 9MG resistance in Plasmodium falciparum laboratory strains 3D7 and Dd2 and identified mutated genes associated with resistance by genome sequencing.
