SRX1225092: RNA-Seq Analysis of Colonial Phase Variants of V. cholerae SD3

Design: RNA samples were chemically fragmented to 400-500 ntds. To create 5´ to 3´ strand-specific cDNA libraries, a TruSeq Stranded Sample Preparation kit was used, followed by gel extraction to select fragments with a minimum size of 80 ntds. Fragments were barcoded and multiplexed with other samples from this study on a single lane of an Illumina HiSeq2000 for 101 cycles. From RNA population of 80-500 ntds, 100 ntds were sequenced from randomly selected ends using the TruSeq SBS Sequencing Kit Version 3 and demultiplexed with Casava 1.8.2.
