SRX26886989: RNA-Seq of Arabidopsis thaliana: guard cell nuclear RNA
1 ILLUMINA (NextSeq 500) run: 36.3M spots, 2.8G bases, 1.1Gb downloads

Design: Nuclei were isolated using a modified INTACT protocol. RNA was extracted from nuclei using proteinase K treatment, followed by Trizol extraction and RNeasy micro columns. RNA quality was assessed using Bioanalyzer and Qubit. Libraries were prepared using TruSeq Stranded Total Plant RNA with Ribo-Zero Plant, employing rRNA depletion. Sequencing was performed on NextSeq500, generating 30 million 75bp reads per sample. Samples were sequenced in triplicate, except KAT1:INTACT at 40% SWC (duplicate) and 20% SWC (quadruplicate).

The study aimed to generate guard cell-specific RNA-seq transcriptomes in Arabidopsis thaliana during progressive drought stress and rewatering, while optimizing the INTACT protocol for RNA preservation in leaf tissue. By comparing gene expression changes between guard cells and bulk leaf tissue, this research enhances our understanding of guard cell-specific responses to drought, identifies unique and shared expression patterns, and provides valuable transcriptomic data that can inform strategies for improving drought tolerance and water use efficiency in crops.
