Design: Two hundred to 500 mg of dried leaf tissue was homogenized using a FastPrep-24TM 5G bead beating and lysis system (MP Biomedicals, 237 Solon, Ohio, United States). DNA extraction followed a modified sorbitol extraction protocol (torchov et al. 2000). Double-stranded DNA concentration was quantified using a Qubit 4 Fluorometer (Invitrogen, Waltham, Massachusetts, United States). Fragment size was assessed on a 2% agarose gel. Extractions from samples with low DNA concentrations were repeated and subsequently pooled to obtain a sufficient DNA quantity (250 ng) for successful library preparation. However, twelve successfully sequenced samples were below this threshold. Library preparation, target enrichment, and sequencing of 150-bp paired-end reads were carried out at RAPiD Genomics LLC (Gainesville, Florida, USA) using an Illumina HiSeq 3000 (Illumina Inc., San Diego, California, USA). Samples with a high concentration of high molecular weight DNA were sheared during library preparation. Shearing of DNA extracts with degraded DNA (as confirmed by gel electrophoresis) was bypassed. A probe set specifically designed to target 745 low-copy nuclear loci within Burmeistera, a closely related genus, was used for target enrichment (Bageley et al., 2020) and 150 bp paired-end reads were sequenced in an Illumina HiSeq 3000.
