Glucocorticoid and Mineralocorticoid Receptor Signaling have Sex-dependent and Independent Effects in Mouse Hippocampus

(Submitter supplied) Many stress-related neuropsychiatric disorders display pronounced sex differences in their frequency and clinical symptoms. Glucocorticoids are primary stress hormones that have been implicated in the development of these disorders but whether they contribute to the observed sex bias is poorly understood. Glucocorticoids signal through two closely related nuclear receptors, the glucocorticoid (GR) and mineralocorticoid receptor (MR), that are both expressed at high levels in the hippocampus. more...

Genome activity during the emergence of mouse embryonic totipotency (time course)

(Submitter supplied) Fertilization transforms sperm and egg into a totipotent embryo but the underlying mechanisms are unknown. We here report that gene expression initiates during the gamete-to-embryo transition in mouse embryos. Meiotic exit induced by sperm entry enhances a transcriptionally-permissive epigenetic landscape. Time-course analysis of single embryos revealed a succession of genome-wide transcription 'ripples' initiating within 2 hours. more...

Transcriptome profiling of left ventricles from Wildtype and KDM5B knockout mice

(Submitter supplied) Myocardial fibrosis is the most common pathological feather of adverse ventricular remodeling, and persistent fibrotic extension decreases myocardial compliance, promotes the development of heart failure. Epigenetics has been considered to play a potent regulatory role in the development of excessive myocardial fibrosis. Although, the explicit mechanism of epigenetic regulation in myocardial fibrosis still needs to be fully elucidated.Transcriptome microarray analysis was used to screen differentially expressed genes in myocardial tissues from KDM5B KO and littermate control WT mice at day 7 after MI operation.

Ethanol and its nonoxidative metabolites promote acute liver injury by inducing ER stress, adipocyte death and lipolysis

(Submitter supplied) We gave mice either a 5g/kg ethanol (EtOH) gavage, a 7g/kg EtOH gavage, or an isocaloric maltose gavage (equivalent to 5g/kg EtOH) in the morning and sacrificed mice 9h later. Microarray analyses were performed on liver tissue from these mice to determine genes and pathways contributing to acute ethanol-induced liver injury. Lipid metabolism-associated genes were found to be dysregulated in both the 5g/kg and 7g/kg groups, with the majority of changes occuring in the 7g/kg group. more...

Hepatic CYP2B10 is highly induced by binge ethanol and contributes to acute-on-chronic alcohol-induced liver injury

(Submitter supplied) We altered the NIAAA (Gao Binge Model) of 8 week chronic plus binge ethanol (E8G1) to have 3 total ethanol gavages (5 g/kg; 1 every 24h) of ethanol after 8 weeks of chronic ethanol feeding (E8G3) to determine how the liver responds to chronic plus consecutive binge ethanol. Surprisingly, E8G3 treatment induced lower levels of liver injury, steatosis, inflammation, and fibrosis as compared to mice after E8G1 treatment. more...

IκBζ-deficient epidermis mediates systemic autoimmune inflammation via skin dysbiosis.

(Submitter supplied) Skin microbiota affect systemic inflammation through mechanisms that have not been completely elucidated. We previously demonstrated that keratinocyte-specific IκBζ-deficient mice spontaneously develop autoimmune inflammation resembling human Sjögren syndrome. In this study, we examined how IκBζ-deficient epidermis dictates systemic autoimmune inflammation onset. To examine if IκBζ-deficient keratinocytes are susceptible to apoptotic stimuli in a steady state, we performed microarray analysis of untreated murine back epidermis. more...

An inactivated multivalent influenza A virus vaccine is broadly protective in mice and ferrets [mouse]

(Submitter supplied) Influenza A viruses (IAVs) present major public health threats from annual seasonal epidemics and pandemics as well as from viruses adapted to a variety of animals including poultry, pigs, and horses. Vaccines that broadly protect against all such IAVs, so-called “universal” influenza vaccines, do not currently exist, but are urgently needed. Here, we demonstrated that an inactivated, multivalent whole virus vaccine, delivered intramuscularly or intranasally, was broadly protective against challenges with multiple IAV hemagglutinin and neuraminidase subtypes in both mice and ferrets. more...

Altered transcriptional responses in the lungs of aged mice after influenza infection

(Submitter supplied) This study used transcriptomic analysis to evaluate how aging specifically affects the functional host response to influenza in the mouse lung. Adult (12-16 weeks) and aged (72-76 weeks) female BALB/c mice were infected with influenza (A/Puerto Rico/8/34 (H1N1) (A/PR8)) intranasally (instilled with 50 μl of PBS containing 50-100 pfu PR8). Lungs were processed for RNA expression analysis via Agilent microarray at various time points. more...
