Whole Genome Association Study on Tissue Tropism Phenotypes in Group A Streptococcus

(Submitter supplied) Group A Streptococcus (GAS) has a rich evolutionary history of horizontal transfer among its core genes. Yet, despite extensive genetic mixing, GAS strains have discrete ecological phenotypes. To further our understanding of the molecular basis for ecological phenotypes, comparative genomic hybridization of a set of 97 diverse strains to a GAS pan-genome microarray was undertaken, and the association of accessory genes with emm genotypes that define tissue tropisms for infection was determined. more...

Comparison of the CcpA and CovR Transcriptomes in Group A Streptococcus

(Submitter supplied) The goal of this study were to compare the transcriptoms of CcpA and CovR in group A Streptococcus. Isogenic mutant strains were created in which CcpA and CovR were inactivated alone and in combination. The transcriptomes of the four strains (wild-type, CcpA mutant, CovR mutant, and ccpA-covR double mutant) were then compared.

Determination of transcriptome of SptR in group A Streptococcus during growth in human saliva

(Submitter supplied) The molecular genetic mechanisms used by bacteria to persist in humans are poorly understood. Group A Streptococcus (GAS) causes the majority of bacterial pharyngitis cases in humans and is prone to persistently inhabit the upper respiratory tract. To gain information about how GAS survives in and infects the oropharynx, we analyzed the transcriptome of a serotype M1 strain grown in saliva. The dynamic pattern of changes in transcripts of genes [spy0874/0875, herein named sptR and sptS (sptR/S), for saliva persistence] encoding a two-component gene regulatory system of unknown function suggested that SptR/S contributed to persistence of GAS in saliva. more...

Expression comparison of GAS M3 strains MGAS315 & MGAS9887 in exponential growth

(Submitter supplied) Epidemiological data indicate that strains MGAS315 and MGAS9887 differ significantly in their capacity to cause necrotizing fasciitis infections. To probe the hypothesis that this difference was associated with differences in gene expression, an expression MA comparison of the strains in exponential phase growth in rich media was conducted. Streptococcus pyogenes (Group A Strep) amplicon array

(Submitter supplied) Pan-genome microarray was constructed using Alab49 as the reference. PCR primers were designed for predicted genes in Alab49 using Primer3, followed by 11 subsequent strains not already represented by Alab49 and were tested for uniqueness using in-house scripts. The resulting 3388 amplicons ranged in size from 51 to 1170 bp; all of these were amplified using 10X PCR Buffer, MgCl2, 100mM dNTP mix, Taq Polymerase, and water (all Applied Biosystems). more...
