Pathway and single gene analysis of Caco-2 cell differentiation by ascorbate-stabilized quercetin

(Submitter supplied) The aim was to investigate mechanisms contributing to quercetin’s previously described effects on cell-proliferation and -differentiation, which contradicted its proposed anti-carcinogenic potency. In a 10-day experiment, 40 µM quercetin stabilized by 1mM ascorbate reduced Caco-2 differentiation up to 50% (P<0.001). Caco-2 RNA from days 5 and 10, hybridized on HG-U133A2.0 Affymetrix® GeneChips®, showed 1,743 affected genes on both days (P<0.01). more...

Quercetin effect on intestinal cell differentiation in vitro: time course

Analysis of post-confluent Caco-2 colon cancer cells up to 10 days after treatment with ascorbate-stabilized quercetin, a polyphenol antioxidant compound. Caco-2 cells differentiate after reaching confluency. Results provide insight into quercetin's role in cell-proliferation and -differentiation.

(Submitter supplied) This series of microarray experiments monitored the gene expression profiles for monoclonal cell lines (derived from HEK-293 parental cell culture) with high (H1, H15, H24, H36, H39) or low (L3, L28, L29) levels of store-operated Ca2+ entry (SOCE). For selection of clones, HEK-293 cells were loaded with indo-1 and sorted by FACS on the basis of their cyclopiazonic acid (CPA)-stimulated Ca2+ entry.
