Inflammatory response to titanium dioxide particles exposure is enhanced during pregnancy

(Submitter supplied) Rationale: Maternal immune responses can promote allergy development in offspring. Pilot data show that neonates of mother mice exposed during pregnancy to air pollution particles have increased allergic susceptibility. Objective: We investigated whether inflammatory response to titanium dioxide (TiO2) particles earlier considered immunologically ‘inert’ is enhanced during pregnancy. Methods: Pregnant BALB/c mice (or non-pregnant controls) received particle suspensions intranasally at day 14 of pregnancy. more...

Altered Genomic Expression of Immune and Metabolic Pathways in a Murine Model of Diesel Enhanced Allergic Sensitization

(Submitter supplied) Diesel exhaust (DE) has been shown to enhance allergic sensitization in animals following high dose instillation or chronic inhalation exposure scenarios. The purpose of this study was to determine if short term exposures to diluted DE enhance allergic immune responses to antigen, and identify possible mechanisms using microarray technology. BALB/c mice were exposed to filtered air or diluted DE to yield particle concentrations of 500 or 2000 µg/m3 4 hr/day on days 0-4. more...

In vitro transformation of human bronchial epithelial cells by diesel exhaust particles

(Submitter supplied) The potential of diesel exhaust particles (DEP) to transform human bronchial epithelial cells (HBEC3) was investigated and a stably transformed cell line (T2-HBEC3) was established. Short-term DEP exposure experiments adds information of immunomodulatory effect markers and differences in susceptibility between normal and sensitized bronhial epithelial cells of the human lung.

Macrophage samples from the HMDP

(Submitter supplied) Identify genes involved in regulation of inflammatory responses and gene-environemnt interactions, in macrophages from a set of mouse inbred strains termed the HMDP. The HMDP is a genetically diverse mapping panel comprised of classical inbred and recombinant inbred wild type mice. The RMA values of genes were used for genome wide association as described in Bennett et al Genome Research 2010.

Combined effects of in utero and adult second-hand smoke exposure on adult lungs

Analysis of lung from 15-wk-old males exposed in utero and then as adults to second hand smoke (SHS). These animals exhibit increased airway hyperresponsiveness. Results provide insight into the role of in utero SHS exposure in potentiating subsequent adult physiological responses to SHS exposure.

In utero exposure to diesel exhaust particulates is associated with alterations of the neonatal cardiomyocyte transcriptome, metabolic parturbation and a global decrease in DNA methylation. [RRBS]

(Submitter supplied) Developmental exposure to particulate matter air pollution is harmful to cardiovascular health, but the mechanisms by which this exposure mediates susceptibility to heart disease is poorly understood. We have previously shown, in a mouse model, that gestational exposure to diesel exhaust results in increased cardiac hypertrophy, fibrosis and susceptibility to heart failure in the adult offspring following transverse aortic constriction. more...

In utero exposure to diesel exhaust particulates is associated with an altered cardiac transcriptional response to transverse aortic constriction and altered DNA methylation.

(Submitter supplied) In utero exposure to diesel exhaust particulates has been associated with increased adult susceptibility to heart failure in mice but the mechanisms by which this exposure promotes susceptibility are poorly understood. To identify potential transcriptional effects that mediate this susceptibility, we have performed RNA-seq analysis on adult hearts from mice exposed to diesel exhaust in utero and that have subsequently undergone transverse aortic constriction. more...

HMEC nine-condition expression profiling

(Submitter supplied) Exposure to air particulate matter is associated with increased cardiovascular morbidity and mortality. One of the possible mechanisms is by promotion of atherosclerosis mediated by synergy of particulate matter chemicals and oxidized phospholipids on vascular endothelial cells. We treated human microvascular endothelial cells (HMEC) in triplicate wells with diesel exhaust particles (DEP) extract at a concentration of 5 and 25 mcg/ml, oxidized 1-palmitoyl-2-arachidonyl-sn-glycero-3-phosphorylcholine (ox-PAPC) at concentrations of 10, 20 and 40 mcg/ml or combination of DEP at 5 mcg/ml plus ox-PAPC at 10, 20 and 40 mcg/ml. more...

Differential Transcriptional Changes in Mice Exposed to Chemically Distinct Diesel Samples

(Submitter supplied) Epidemiological studies have linked exposure to ambient particulate matter (PM) with increased asthmatic symptoms. Diesel exhaust particles (DEP) are a predominant source of vehicle derived ambient PM, and experimental studies have demonstrated that they may have adjuvant potential when given with an antigen. We previously compared 3 DEP samples: N-DEP, A-DEP, and C-DEP in a murine ovalbumin (OVA) mucosal sensitization model and reported the adjuvant activity to be: C-DEP ≈ A-DEP > N-DEP. more...
