Gene expression analysis of embryonic stem cells expressing VE-cadherin (CD144) during endothelial differentiation

(Submitter supplied) Endothelial differentiation occurs during normal vascular development in the developing embryo. Mouse embryonic stem (ES) cells were used to further define the molecular mechanisms of endothelial differentiation. By flow cytometry a population of VEGF-R2 positive cells was identified as early as 2.5 days after differentiation of ES cells, and a subset of VEGF-R2 + cells, that were CD41+ positive at 3.5 days. more...

Transcriptomic analysis of undifferentiated hESC, hESC -derived cell populations and human Embryonic Liver (EL) cell populations.

(Submitter supplied) RNA microarrays technology was used to compare hESC-derived cell populations to undifferentiated hESC and to human EL cell populations. Microarrays confirmed the hematoendothelial, endothelial and hematopoietic identity of hPSC-derived CD144+-EBs, BCs and ECs, respectively, and the similarities between hESC-derived cell populations and human EL equivalent populations.

Early embryoid body differentiation

(Submitter supplied) Embryonic stem (ES) cells, when grown in suspension culture without feeders, spontaneously form round structures known as embryoid bodies. Given the appropriate conditions, these cells can differentiate over time into precursors of all three germ layers. Embryoid bodies, in a disorganized way, mimic early embryonic development to a certain extent and can be used as a synchronously differentiating large scale source of tissue for the study of biological determinants of early differentiation. more...

Comparison of undifferentiated ES cell lines HM1, IMT11, SHBL6.3

(Submitter supplied) 3 different ES cell lines were compared in order to determine whether there are significant expression profile differences between ES cell lines, or whether the constraints of maintaining pluripotency in culture force a similar expression profile on cell lines derived from disparate sources. Our results indicate that the latter is more likely. We identified 21 genes that were significantly differentially regulated, either on comparison with the pooled control, or on direct comparison of individual ES cell line data from different slides. more...

Role of miRNAs in the differentiation of human embryonic stem cells to an endothelial cell lineage

(Submitter supplied) To assess changes in the miRNA-ome during the early stages of endothelial differentiation, the expression of mature human miRNAs at days 2, 4 and 10 of endothelial differentiation (vs pluripotent time-matched samples) in SA461 hESC cell line was analysed in a two-channel microarray. Expression of the pluripotency-associated miRNAs miR-302a – d and miR-372 and miR-373 was significantly suppressed with progression of differentiation. more...

Transcriptional Characterization of Human Embryonic Stem Cell-Derived Endothelial Cells

(Submitter supplied) Differentiation of human embryonic stem cells into endothelial cells (hESC-ECs) has the potential to provide an unlimited source of cells for novel transplantation therapies of ischemic diseases by supporting angiogenesis and vasculogenesis. In this study, we developed an extracellular matrix culture system for increasing endothelial differentiation and free from contaminating animal cells. We investigated the transcriptional changes that occur during endothelial differentiation of hESCs using whole genome microarray, and compared to human umbilical vein endothelial cells (HUVECs). more...

Expression data from differentiating Flk1- and Flk1+ ES cells expressing Snail during Wnt inhibition

(Submitter supplied) ES cells differentiated in the presence of the Wnt inhibitor DKK1 fail to express the transcription factor Snail and undergo EMT or mesoderm differentiation. We generated an ES cell line, A2.snail, that induced Snail expression upon addition of doxycycline addition. Microarrays were used to gain a global picture of Flk1- and Flk1+ cells generated one day after Snail was expressed during Wnt inhibition.

Human Pluripotent Cell Derived Endothelial Cell Characterization

(Submitter supplied) Human ES or iPS Cells were differentiated into endothelial cells (ECs) defined by expression of CD31 (PECAM1) and CD144 (VE-Cadherin) on the cell surface. All ES or iPS derived ECs were greater than 90% double positive for these two markers. These in vitro derived ECs were compared to each other and to ECs from primary cell sources; arterial (aortic Ecs), venous (saphenous vein) and lymphatic.

Comparison of human embroynic stem cell derived vascular cells to mature human vascular and hematopoietic cells

(Submitter supplied) The pathways involved in hierarchical differentiation of human embryonic stem cells (hESC) into abundant and durable endothelial cells (EC) are unknown. We employed an EC-specific VE-cadherin promoter driving GFP (hVPr-GFP) to screen for factors that augmented yields of vascular-committed ECs from hESCs. In phase 1 of our approach, inhibition of TGFb, precisely at day 7 of hESC differentiation, enhanced emergence of hVPr-GFP+ ECs by 10-fold. more...

Defining Developmental Potency and Cell Lineage Trajectories by Expression Profiling of Differentiating Mouse ES Cells

(Submitter supplied) Biologists rely on morphology, function, and specific markers to define the differentiation status of cells. Transcript profiling has expanded the repertoire of these markers by providing the snapshot of cellular status that reflects the activity of all genes. However, such data have been used only to assess relative similarities and differences of these cells. Here we show that principal component analysis (PCA) of global gene expression profiles map cells in multidimensional transcript profile space and the positions of differentiating cells progress in a stepwise manner along trajectories starting from undifferentiated embryonic stem (ES) cells located in the apex. more...
