Gene expression data from AP-2γ silenced MCF-7 cells

(Submitter supplied) Overexpression of the AP-2γ transcription factor in breast tumours has been identified as an independent predictor of poor outcome and failure of hormone therapy, even in ER positive, ErbB2 negative tumours; markers of a more favourable prognosis. To understand further the role of AP-2γ in breast carcinoma, we have used an RNA interference and gene expression profiling strategy using the MCF-7 cell line as a model for ER positive, ErbB2 negative tumours with AP-2γ overexpression. more...

Activator protein-2gamma silencing effect on MCF-7 breast tumor cell line

Analysis of MCF-7 cells silenced for activator protein (AP)-2γ transcription factor using three distinct self-targeting sequences (AP-2γ siRNA1-3). AP-2γ overexpression in breast tumors is a predictor of poor outcome. Results provide further insight into the role of AP-2γ in breast carcinoma.

A TFAP2C Gene Signature is Predictive of Outcome in HER2 Breast Cancer (ChIP-Seq)

(Submitter supplied) A subset of HER2 breast cancers with amplification of the TFAP2C gene locus becomes addicted to AP-2g. We sought to define AP-2g-regulated genes that control growth and invasiveness by comparing HER2 cell lines with differential response to TFAP2C knockdown. A set of 68 differentially expressed genes was identified, which included CDH5 and CDKN1A. Pathway analysis implicated the MAPK13/p38δ and retinoic acid regulatory nodes, which were confirmed to display divergent responses. more...

Activator protein-2γ is a critical determinant of estrogen receptor interactome formation and gene transcription in breast cancer [expression]

(Submitter supplied) Estrogen receptor α (ERα) is key player in the progression of breast cancer. ERα binds to DNA and mediates long-range chromatin interactions throughout the genome, but the underlying mechanism in this process is unclear. Here, we show that AP-2 motifs are highly enriched in the ERα binding sites (ERBS) identified from the recent ChIA-PET of ERα. More importantly, we demonstrate that AP-2γ (also known as TFAP2C), a member of the AP-2 family which has been implicated in breast cancer oncogenesis, is recruited to chromatin in a ligand-independent manner and co-localized with ERα binding events. more...

TRIM37 augments AP-2γ transcriptional activity and cellular localization via K63-linked polyubiquitination to drive breast cancer progression [ChIP-seq]

(Submitter supplied) Using a proteomics approach, we identified the Tripartite Motif Containing 37 (TRIM37) as a novel transcriptional coactivator of AP-2γ. We demonstrate TRIM37 facilitates AP-2γ chromatin binding to regulate the AP-2γ mediated transcriptional program directly. We provide evidence that TRIM37 achieves this by stimulating K63-chain-linked polyubiquitination of AP-2γ, promoting protein localization from the cytoplasm to the nucleus. more...

Transgenic overexpression of Tcfap2c/AP-2gamma results in liver steatosis

(Submitter supplied) We established a mouse model, in which transcription factor Tcfap2c can be activated in an inducible and reversible manner in somatic tissues, taking advantage of the tetracycline-dependent regulatory system. Induction of the transgene led to robust expression in all tissues (except brain and testis) and lead to rapid mortality within 3-7 days. In the liver, accumulation of microvesicular lipid droplets and breakdown of major hepatic metabolism pathways resulted in steatosis.
