Expression data from TKI258 treated 4T1 tumors

(Submitter supplied) 4T1 mouse mammary carcinoma cells have an autocrine FGFR active loop leading to constitutive activation of downstream signaling pathways. We found that FGFR inhibitors have a strong effect on 4T1 tumors in-vivo. We used microarray to understand the contribution of FGFR signaling to the tumor formation upon TKI258 treatment.

Epithelial-to-mesenchymal transition defines feedback activation of receptor tyrosine kinase signaling induced by MEK inhibition in KRAS mutant lung cancer

(Submitter supplied) To determine the difference of gene expression profile in epithelial and mesenchymal KRAS mutant lung cancers, epithelial NCI-H358 cells were treated with TGFβ1 (4 ng/mL) or PBS for 14 days in order to induce epithelial to mesenchymal transition (EMT). Gene expression was determined in NCI-H358 cells before and after EMT induction. In addition, in order to investigate the effect of a MEK inhibitor trametinib on gene expression, mesenchymal NCI-H1792 cells were treated with 50 nM trametinib for 48 hours. more...

A Splice Variant of HER2 Activates Key Signaling Cascades and Evokes Mammary Tumors and Metastases

(Submitter supplied) The Epidermal Growth Factor Receptor 2 (ERBB2 or HER2) is amplified and overexpressed in approximately 20% of invasive breast cancers and is associated with metastasis and poor prognosis. Here we describe the role of a constitutively active splice variant of HER2 (Delta-HER2) in human mammary epithelial cells. Overexpression of Delta-HER2 in human mammary cells decreased apoptosis and increased proliferation and expression of epithelial-to-mesenchymal markers. more...

JNK3 null vs. wild type hippocampi

(Submitter supplied) JNK is a member of the mitogen-activated protein kinase (MAPK) family and there are three genes that encode for JNK1, JNK2 and JNK3 respectively. JNK3 is mainly expressed in the central nervous system and it plays a crucial role in neuronal death in several neurodegenerative diseases, including epilepsy, Alzheimer’s disease, Parkinson’s disease and Huntington’s disease. By contrast, the isoforms JNK1 and JNK2 seem to be involved in brain development. more...

Lentiviral shRNA library screen to identify growth-related genes in malignant melanoma

(Submitter supplied) Little is known about genes that promote melanoma cell growth and proliferation. siRNAs may be used to address the role of individual genesin these processes. RNAi library screens were used in the past to gain a comprehensive overview of all genes involved in cell growth, proliferation, migration and other cellular processes. A large-scale loss-of-function screen for eight different melanoma cell lines was performed using a pooled lentiviral shRNA library (GeneNet Human 50K lentiviral shRNA Library,cat#SI206B-1, System Biosciences) to identify genes relevant for melanoma cell growth and proliferation. more...

The Rho Exchange Factors Vav2 and Vav3 Control a Lung Metastasis–Specific Transcriptional Program in Breast Cancer Cells

(Submitter supplied) The guanosine triphosphatases of the Rho and Rac subfamilies regulate protumorigenic pathways and are activated by guanine nucleotide exchange factors (Rho GEFs), which could be potential targets for anticancer therapies. We report that two Rho GEFs, Vav2 and Vav3, play synergistic roles in breast cancer by sustaining tumor growth, neoangiogenesis, and many of the steps involved in lung-specific metastasis. more...

FGFR signature

(Submitter supplied) Drugs that target specific gene alterations have proven beneficial in the treatment of cancer. Because cancer cells have multiple resistance mechanisms, it is important to understand the downstream pathways of the target genes and monitor the pharmacodynamic markers associated with therapeutic efficacy. We performed a transcriptome analysis to characterize the response of various cancer cell lines.

The TLR2 pathway is required for self-renewal of mammary cancer initiating cells

(Submitter supplied) In the past few years, mammary cancer initiating cells (CICs) have been identified in mouse and human as a subpopulation of tumor cells that selectively posses tumor initiation and self-renewal capacity and the ability to give rise to bulk populations of non-tumorigenic cancer cells progeny through differentiation. They could also be responsible for tumor progression, metastasis, resistance to therapy and recurrence. more...

Cytosolic phospholipase A2 alpha regulates TLR signaling and migration in metastatic 4T1 cells

(Submitter supplied) We aimed at characterizing the response to reduced cPLA2α activity in metastatic versus non-metastatic cells. We employed an isogenic murine cell line pair displaying metastatic (4T1) and non-metastatic (67NR) phenotype to investigate the role of cPLA2α on migration. Further, we elucidate the effect of reduced cPLA2α activity on global gene expression in the metastatic cell line We found that cPLA2α inhibition may inhibit metastasis through an anti-migratory effect, possibly involving Toll-like receptor signaling and type I interferons
