CGH array profiles of melanoma cell lines

(Submitter supplied) Dysfunction in type I interferon (IFN) signaling occurs in patients with stage II or more advanced cancer. After screening the effects of a panel of 12 melanoma cell lines on PBMCs of healthy volunteers of IFNalpha signal pathway, two groups of melanoma cell lines could be identified one with stronger suppression (low pSTAT-1 group) than the other (high pSTAT-1 group). Comparative genomic hybridization (CGH) identified consistent amplification of 12q22-24 as a genomic marker for the immune suppressive melanoma cell lines. more...

Comparative gene expression profiles of immune inhibitory and non-inhibitory melanoma cell lines

Comparative global gene expression between two groups identified 6771 differential expression genes. more...

Comparative gene expression profiles of NOS1-KO and WT A375 melanoma cell lines

(Submitter supplied) NOS1 plays a vital role in tumor. A cell model of NOS1 gene knockout in human melanoma cell line A375 was constructed using CRISPR/Cas9 technique for the study of its function. We used microarrays to detail the Global gene expression underlying NOS1-knockout compare to wild type A375 cell.

Expression profiling reveals novel pathways in the transformation of melanocytes to melanomas. II

(Submitter supplied) Spotted oligonucleotide microarrays were used to assess global differential gene expression comparing normal human melanocytes with six independent melanoma cell strains from advanced lesions. The data, validated at the protein level for selected genes, confirmed the overexpression in melanoma cells relative to normal melanocytes of several genes in the growth factor/receptor family that confer growth advantage and metastasis. more...

Melanocyte to melanoma transformation

Expression profiling of normal melanocytes, primary melanoma cells, and metastatic melanoma cells. Results provide insight into changes in advanced melanoma relative to normal melanocytes and reveal new targets that can be used in assessing prognosis, staging, and therapy of melanoma patients.

Suppression of type I interferon signaling overcomes the oncogene-induced senescence and mediates melanoma development and progression

(Submitter supplied) Activation of oncogenes often leads to induction of the DNA damage responses and onset of the cell senescence. Given that DNA damage can also trigger production of type I interferons (IFN) that contribute to senescence development, we sought to determine the role of IFN in the oncogene-induced senescence. Our data in mouse model demonstrate that inactivation of IFN signaling is sufficient for inducing melanomas in melanocytes harboring mutant Braf. more...
