Angiopoietin-like proteins stimulate ex vivo expansion of hematopoietic stem cells

(Submitter supplied) Successful ex vivo expansion of hematopoietic stem cells (HSCs) would greatly benefit the treatment of disease and the understanding of critical questions of stem cell biology. Microarray studies showed that the HSC supportive mouse fetal liver CD3+ cells specifically expressed angiopoietin-like 2 (Angptl2) and angiopoietin-like 3 (Angptl3). When highly enriched HSCs were cultured in the presence of Angptl2 or Angptl3 together with saturating levels of other growth factors for 10 days, a 24 or 30 fold net expansion of long-term HSCs was observed by reconstitution analysis. more...

Inflammatory signaling regulates fetal hematopoiesis

(Submitter supplied) Multiple signaling pathways contribute to blood cell formation in the fetus, but a role for innate immune/inflammatory signaling has not been described. Here we show that mouse fetuses deficient for either interferon gamma 1 (IFNg) or its receptor have decreased numbers of lymphoid progenitors and hematopoietic stem cells (HSCs) in the aorta/gonad/mesonephros region and placenta. The role of IFNg signaling was evolutionarily conserved, as morpholino knockdown of IFNg and its receptor reduced the number of hematopoietic stem and progenitor cells in the dorsal aorta and caudal hematopoietic territory of zebrafish embryos, and rag2 expressing cells in the thymus. more...

Transcriptomics analysis of gene expressions, m6A enrichment levels and ythdf2 binding targets in control and mettl3 or ythdf2 morphants zebrafish embryos

(Submitter supplied) RNA was isolated from control and mettl3 or ythdf2 morphants zebrafish cells using the TRIzol (Invitrogen) reagent by following the company manual. For all samples the RNA integrity was checked using an Agilent Bioanalyzer 2100. All samples showed a RIN (RNA integrity number) of higher than 9. Approximately 2.5 µg of total RNA was then used for library preparation using a TruSeq™ RNA Sample Prep Kit v2 (Illumina, San Diego, CA, USA) according to the manufacturer’s protocol.The libraries were sequenced using HiSeq2500 or HiSeq 3000 (Illumina) in paired-read mode, creating reads with a length of 101 or 151 bp. more...
