A NOVEL EPIGENETIC SILENCING PATHWAY INVOLVING the HIGHLY CONSERVED 5'-3' EXORIBONUCLEASE Dhp1/Rat1/Xrn2 in SCHIZOSACCHAROMYCES POMBE [gene expression]

(Submitter supplied) Epigenetic gene silencing plays a critical role in regulating gene expression and contributes to organismal development and cell fate acquisition in eukaryotes. In fission yeast, Schizosaccharomyces pombe, heterochromatin-associated gene silencing is known to be mediated by RNA processing pathways including RNA interference (RNAi) and a 3’-5’ exoribonuclease complex exosome. Here, we report a new RNA-processing pathway that contributes to epigenetic gene silencing and assembly of heterochromatin mediated by 5’-3’ exoribonuclease Dhp1/Xrn2. more...

A conserved factor Dhp1/Rat1/Xrn2 triggers premature transcription termination and nucleates heterochromatin to promote gene silencing [sRNA-seq]

(Submitter supplied) Co-transcriptional RNA processing and surveillance factors mediate heterochromatin formation in fission yeast. In addition to RNAi, RNA elimination machinery including MTREC (Mtl1-Red1 core) and the exosome are involved in facultative heterochromatin assembly, however, the exact mechanisms remain unclear. Here we show that RNA elimination factors cooperate with the conserved exoribonuclease Dhp1/Rat1/Xrn2, which couples pre-mRNA 3’-end processing to transcription termination, to promote premature termination and facultative heterochromatin formation at meiotic genes. more...

The Chromatin-Remodeling Factor FACT Contributes to Centromeric Heterochromatin Independently of RNAi

(Submitter supplied) Centromeres exert vital cellular functions in mitosis and meiosis. A specialized histone and other chromatin-bound factors nucleate a dynamic protein assembly that is required for the proper segregation of sister chromatids. In several organisms including the fission yeast S. pombe, a RNAi-related pathway further strengthens the epigenetic maintenance of centromere location, primarily through repressing transcription near centromeres. more...

Accumulation of RNA on chromatin disrupts heterochromatic silencing

(Submitter supplied) Long non-coding RNAs (lncRNAs) play a conserved role in regulating gene expression, chromatin dynamics and cell differentiation. They serve as a platform for RNA interference (RNAi)-mediated heterochromatin formation or DNA methylation in many eukaryotic organisms. We found in Schizosaccharomyces pombe, that heterochromatin is lost at transcribed regions in absence of RNA degradation, although establishment is not affected. more...

Nuclear aconitase regulates heterochromatin formation by interacting with Chp1

(Submitter supplied) Aconitase is a highly conserved metabolic enzyme for TCA cycle. In Schizosaccharomyces pombe, an aconitase (Aco2) is localized in both mitochondria and the nucleus, apart from an exclusively mitochondrial one (Aco1). We investigated the role of nuclear Aco2, and present evidences that it interacts with Chp1, a heterochromatin-associated HP1 protein, and acts as an anti-silencing factor. Nuclear depletion of Aco2 by deleting nuclear localization signal (aco2ΔNLS) suppressed the gene-silencing defects of RNAi mutants Δago1 and Δdcr1 at the centromere, where RNAi-mediated heterochromatin formation prevails. more...

An RNA Endonuclease-Kinase Complex Required for Spreading and Epigenetic Inheritance of Heterochromatin

(Submitter supplied) The highly conserved multienzyme Rix1-containing complex (hereafter referred to as the rixosome), is required for ribosomal RNA (rRNA) processing and also localizes to heterochromatin in fission yeast, but its role in heterochromatin formation is unknown. Here we report the isolation of separation of function mutations in subunits of the rixosome that abolish its physical association with Swi6/HP1 and localization to heterochromatin, but do not affect growth or rRNA processing. more...

Argonaute and Trimmer generate Dicer-independent priRNAs and nucleate RNAi

(Submitter supplied) RNA interference is involved in silencing of transposable and repetitive elements. How these elements are initially recognized by RNAi is a fundamental and unanswered question. We previously identified a class of Dicer-independent small RNAs, called primal small RNAs (priRNAs), in fission yeast. The mechanism by which Dicer-independent small RNAs are generated is not clear for any species. Here we reconstitute priRNA biogenesis in vitro and demonstrate that priRNAs can nucleate RNAi in vivo. more...

Genome-wide mapping of Ccp1 association sites in fission yeast

(Submitter supplied) In our genetic screen, we found that Ccp1 is an epigenetic stability modulator in centromeres and heterochromatin in S. pombe. In interphase cells,Ccp1-GFP enriched at centromeres. To determine the association sites of Ccp1 with the centromeres and to characterize the global distribution pattern of Ccp1 throughout the genome, ChIP-seq were performed for Myc-tagged Ccp1 (C-terminal fusion protein). The result revealed that Ccp1 associates with the central core regions (cnt and imr) within every single centromere.

The Conserved RNA Binding Cyclophilin, Rct1, Regulates Small RNA Biogenesis and Splicing Independent of Heterochromatin Assembly [small RNA]

(Submitter supplied) RNAi factors and their catalytic activities are essential for heterochromatin assembly in S. pombe. This has led to the idea that siRNAs can promote H3K9 methylation by recruiting the Cryptic Loci Regulator Complex (CLRC), also known as Recombination In K Complex (RIKC), to the nucleation site. The conserved RNA-binding protein Rct1 (AtCyp59/SIG-7) interacts with splicing factors and with RNA polymerase II. more...
