Gene expression profiling of in vitro matured bovine oocytes cultivated with or without cumulus cells.

(Submitter supplied) Cumulus cells, surrounding the oocyte, play a key role in the acquisition of oocyte competence to be fertilized and to sustain early embryo development. Cumulus cells contribute to oocyte development by metabolizing energy substrates such as glutathione that may protect the oocyte from oxidative stress damages. The aim of our study was to compare transcriptomics profiles of cumulus enclosed (CEO) and cumulus denuded (CDO) oocytes after in vitro maturation.

Bovine adult versus prepubertal cumulus cells

(Submitter supplied) The present study was undertaken to discover molecular markers in bovine cumulus cells predictive of oocyte competence and elucidate their functional significance. Differences in RNA transcript abundance in cumulus cells harvested from oocytes of adult versus prepubertal animals (model of poor oocyte quality) were identified by microarray analysis. Four genes of interest encoding for the lysosomal cysteine proteinases cathepsin B, S, K and Z and displaying greater transcript abundance in cumulus cells surrounding oocytes harvested from prepubertal animals were chosen for further investigation. more...

Functional signaling and gene regulatory networks between the oocyte and the surrounding cumulus cells

(Submitter supplied) The maturation of an oocyte, the female gamete, during folliculogenesis is highly dependent on the molecular interaction with the somatic cells. Here we generated transcriptome data of single oocytes and the surrounding cumulus cells from antral follicles, and developed an analytical framework to unveil their inter-cellular communication. We identified hundreds of ligand-receptors pairs that can transduce paracrine signaling between oocyte and cumulus. more...

Extensive effects of in vitro oocyte maturation on rhesus monkey cumulus cell transcriptome

(Submitter supplied) The elaboration of a quality oocyte is integrally linked to the correct developmental progression of cumulus cell phenotype. In humans and non-human primates, oocyte quality is diminished with in vitro maturation. To determine the changes in gene expression in rhesus monkey cumulus cells (CC) that occur during the final day prior to oocyte maturation and how these changes differ between in vitro and in vivo maturation (IVM and VVM), we completed a detailed comparison of transcriptomes using the Affymetrix gene array. more...

RNA‐sequencing reveals genes linked with oocyte developmental potential in bovine cumulus cells

(Submitter supplied) Cumulus cells provide an interesting biological material to perform analyses to understand the molecular clues determining oocyte competence. The objective of this study was to analyze the transcriptional differences between cumulus cells from oocytes exhibiting different developmental potentials following individual in vitro embryo production by RNA‐seq. Cumulus cells were allocated into three groups according to the developmental potential of the oocyte following fertilization: (1) oocytes developing to blastocysts (Bl+), (2) oocytes cleaving but arresting development before the blastocyst stage (Bl-), and (3) oocytes not cleaving (Cl-).

Pyruvate metabolism alters the epigenetic and transcriptional profile of bovine oocytes.

(Submitter supplied) Pyruvate, the end-product of glycolysis in aerobic conditions, is converted into acetyl-CoA inside the mitochondria of oocytes as a master fuel input for the tricarboxylic acid (TCA) cycle. The citrate generated in the TCA cycle can be directed to the cytoplasm and converted back to acetyl-CoA, being driven to lipid synthesis or, alternatively, being used as the substrate for histones acetylation. This work aimed to verify the impact of pyruvate metabolism on the dynamics of lysine 9 histone 3 acetylation (H3K9ac) and RNA transcription in bovine oocytes during in vitro maturation (IVM). more...
