Methylomic Analysis reveals Epigenetic Pattern of SH2B1 in Chinese Monozygotic Twins Discordant for Autism Spectrum Disorder [Bisulfite-seq]

(Submitter supplied) Autism spectrum disorder(ASD) is a complex neurodevelopmental disorder. Aberrant DNA methylation has been observed in ASD but the mechanisms remain largely unknown. Here, we employed discordant monozygotic twins to investigate the contribution of DNA methylation to ASD etiology. Genome-wide DNA methylation analysis was performed using samples obtained from five pairs of ASD-discordant monozygotic twins, which revealed a total of 2397 differentially methylated genes. more...

DNA Methylation Profiling of Human Saliva

(Submitter supplied) Background: Low birth weight is associated with an increased adult metabolic disease risk. It is widely discussed that poor intrauterine conditions could induce long-lasting epigenetic modifications, leading to systemic changes in regulation of metabolic genes. In a unique cohort of 17 monozygotic (MZ) monochorionic female twins very discordant for birth weight (relative differences ranging from 21.3-35.7%), we examined if adverse prenatal growth conditions experienced by the smaller co-twins lead to systemic long-lasting DNA methylation changes. more...

Genome-wide DNA methylation data of 2 pairs of monozygotic twins clinically discordant for IgA nephropathy

(Submitter supplied) Immunoglobulin A nephropathy (IgAN) is the most common primary glomerulonephritis involves both genetic and environmental factors. DNA methylation, the most studied epigenetic modification, was shown to play a role in IgAN. In two pairs of IgAN-discordant monozygotic (MZ) twins, here we assessed genome-wide DNA methylation profiling and gene expression profiling in order to characterize DNA methylation changes in IgAN and their potential influences on gene expression.

Global methylation profiling of lymphoblastoid cell lines reveals epigenetic contributions to autism spectrum disorders

(Submitter supplied) Autism is currently considered a multigene disorder with epigenetic influences. To investigate the contribution of DNA methylation to autism spectrum disorders, we have recently completed large-scale methylation profiling by CpG island microarray analysis of lymphoblastoid cell lines (LCL) derived from monozygotic twins discordant for diagnosis of autism and their nonautistic siblings. Methylation profiling revealed many candidate genes differentially methylated between discordant MZ twins as well as between both twins and nonautistic siblings. more...

Gene expression profiling of lymphoblastoid cell lines from monozygotic twins discordant in severity of autism

(Submitter supplied) Background: The autism spectrum includes a set of complex multigenic developmental disorders that severely impact the development of language, non-verbal communication, and social skills, and are associated with odd, stereotyped, repetitive behavior and restricted interests. To date, diagnosis of these neurologically based disorders relies predominantly upon behavioral observations often prompted by delayed speech or aberrant behavior, and there are no known genes that can serve as definitive biomarkers for the disorders. more...

Genome-wide DNA methylation analysis in peripheral blood of monozygotic twins informative for psychopathology

(Submitter supplied) Peripheral blood from thirty-four monozygotic twin subjects from the general population (n = 17 twin pairs) was collected for epigenomic analysis via Illumina Infinium HumanMethylation450 Beadchip. All subjects were screened for DSM-IV based criteria for both current and lifetime psychiatric disorders. Out of 17 twin pairs, there were: 7 healthy twin pairs where none of the twins of a pair met criteria for any DSM-IV disorder; 6 discordant twin pairs where only one of the twins of each pair met diagnostic criteria; and 4 concordant twin pairs where both twins of a pair met clinical DSM-IV based criteria.

Analysis of genome-wide methylation of phenotypically discordant monozygotic twins

(Submitter supplied) Human intelligence demonstrates one of the highest heritabilities among human quantitative traits. Phenotypically discordant monozygotic twins provide a way to identify loci reponsible for the phenotypical differences. We performed comprehensive DNA methylation analysis in monozygotic twins manifesting differences in IQ scores. Genes with significantly different methylation status are considered as candidates related to human intelligence.
