The in vitro influence of adiponectin on the porcine endometrium on days 15 to 16 of gestation.

(Submitter supplied) Adiponectin is a hormone secreted primarily by white adipose tissue. Recent studies have shown that adiponectin and its receptors (AdipoR1 and AdipoR2) are expressed in different reproductive tissues, including the ovary and uterus. This newly discovered endocrine system plays an important role in the regulation of reproductive processes. The aim of this study was to explore the influence of adiponectin on global gene expression in the porcine endometrium during early gestation, on days 15 to 16 of pregnancy (implantation period).

Deep sequencing of the porcine endometrial transcriptome on day 12 of pregnancy

(Submitter supplied) The porcine conceptus undergoes rapid differentiation and expansion of its trophoblastic membranes between Days 11 and 12 of gestation. Concomitant with trophoblast elongation, production of conceptus estrogen, the porcine embryonic pregnancy recognition signal increases. Conceptus attachment to the uterine surface epithelium starts after Day 13 initiating epitheliochorial placentation. To analyze the transcriptome changes in the endometrium in the course of maternal recognition of pregnancy (MRP), deep sequencing of endometrial RNA samples of Day 12 pregnant animals (n=4) and corresponding non-pregnant controls (n=4) was performed using Illumina RNA-Seq. more...

Endometrial gene expression profiling in pregnant Meishan and Yorkshire pigs on day 12 of gestation

(Submitter supplied) BACKGROUND: Litter size in pigs is a major factor affecting the profitability in the pig industry. The peri-implantation window in pigs is characterized by the coordinated interactions between the maternal uterine endometrium and the rapidly elongating conceptuses and represents a period of time during which a large percentage of the developing conceptuses are lost. However, the gene expression and regulatory networks in the endometrium contributing to the establishment of the maternal: placental interface remain poorly understood. more...

Differential expression of mRNAs in porcine endometrium

(Submitter supplied) The study allowed for the determination of the mRNAs that were differentially expressed in porcine endometrium on different gestational day and breed. The majority of the mRNAs were differentially expressed during the implantation stage. This study will provide the information to better understanding of the role of these differently expressed mRNAs in the porcine endometrium during pregnancy. Affymetrix mRNA arrays were used to generate mRNA profiles of on the endometrium of Meishan and Yorkshire pigs on days 15 (implantation stage), 26 (post-implantation stage) and 50 (mid-gestation stage) of gestation.

Transcriptomic analyis of the response of porcine PBMC to stimulation with concanavalin A

(Submitter supplied) One of the top priorities of the swine industry worldwide is to decrease the susceptibility of its animals to infectious diseases. There is therefore now an increased focus on the production of more “robust” animals, which have lower susceptibility to a broad range of infectious diseases. One general measure of immune competence is the proliferative response of peripheral blood mononuclear cells (PBMC) to mitogenic stimulation. more...

Transcriptome Analysis of the Endometrium from Chinese Erhualian sows that differ in Calcium Ion Concentration and Litter Size

(Submitter supplied) Embryonic survival rate is an important factor for the fecundity of sows, and it is affected by endometrium secreting histotroph. A higher concentration of calcium ion has been observed in the uterus of the high prolific Erhualian sows (EH) compared with low prolific ones (EL), which suggests EH sows had a greater establishment and maintenance of pregnancy, and thus increase embryonic survival rate during peri-implantation period. more...

Genome-wide analysis of the transcriptional response to porcine reproductive and respiratory syndrome virus infection at the maternal/fetal interface and in the fetus

(Submitter supplied) Porcine Reproductive and Respiratory Syndrome Virus (PRRSV) infection of 3rd trimester pregnant pigs can result in transmission of the virus to the fetus and ultimately death in utero or postnatally. Little is known about the immune response to infection at the maternal-fetal interface and in the fetus itself, or the molecular events behind virus transmission and disease progression in the fetus. To investigate these processes, RNA-sequencing of two tissues, uterine endothelium adjacent to the umbilical attachment site and fetal thymus, was performed 21 days post challenge on four groups of fetuses selected from a large PRRSV challenge experiment of pregnant gilts.

Discovery of candidate genes and pathways in the endometrium regulating ovine blastocyst growth and conceptus elongation

(Submitter supplied) Establishment of pregnancy in ruminants requires blastocyst growth to form an elongated conceptus that produces interferon tau, the pregnancy recognition signal, and initiates implantation. Blastocyst growth and development requires secretions from the uterine endometrium. An early increase in circulating concentrations of progesterone (P4) stimulates blastocyst growth and elongation in ruminants. This study utilized sheep as a model to identify candidate genes and regulatory networks in the endometrium that govern pre-implantation blastocyst growth and development. more...

microRNA expression profile in peri-implantation porcine embryos

(Submitter supplied) Recently, microRNAs (miRNAs) have emerged as new players in the fine tuning of embryo development and implantation in mammals via posttranscriptional gene regulation mechanisms. Applying custom made multispecies arrays we aimed to analyze expression profile of microRNAs in peri-implantation porcine conceptuses/trophoblasts to identify their potential role at the maternal-fetal interface during the critical period of maternal recognition of pregnancy and implantation.

microRNAs and isomiRs repertoire in poricne endometrium revealed by Illumina sequencing

(Submitter supplied) Recently, microRNAs (miRNAs), known to modulate gene expression through post-transcriptional mechanisms, were implied in regulation of early pregnancy events including maternal recognition of pregnancy and implantation. To characterize complex transcriptomic changes in expression of miRNAs in pregnant and cyclic endometria collected on days 12, 16 and 20 was analyzed using Illumina deep sequencing. This research showed repertoire of pregnancy-related miRNAs in porcine endometrium during initial stages of conceptus implantation and during the estrous cycle and sheds light on miRNA-mediated gene expression regulation mechanism at the maternal-conceptus interface.

Messenger RNA profiling of bovine endometrium during the estrous cycle using a custom-made cDNA array

(Submitter supplied) The endometrium provides optimal conditions for the transport of sperm to the oviduct, to the site of fertilization, and later on for the reception of the embryo. To study these changes on the level of gene expression, a messenger RNA expression profiling of endometrium tissue samples collected from 19 cyclic heifers at five stages of the estrous cycle (days 0, 3.5, 12, 18, 20) was performed. RNA was extracted from these tissue samples and analyzed with a custom-made bovine oviduct and endometrium (BOE) cDNA array. more...

Expression data from estrogen disrupted endometrium

(Submitter supplied) Placentation of the conceptus to the surface epithelium is governed through a tightly regulated temporal and spatial window. Premature exogenous steriod exposure causes a shift in the maternal tissue's receptivity and prevents proper placentation. We used Affymetrix microarrays to detail the aberrant changes in uterine gene expression during this window, while removing all conceptus contribution to this dysfunction.
