Lymphatic endothelial cell response to Prox1 and NR2F2 depletion

Analysis of lymphatic endothelial cells (LECs) depleted for Prox1, NR2F2, or both regulators. LECs are derived from venous endothelial cells (VECs). Prox1 and NR2F2 regulate LEC and VEC development, respectively. Results provide insight into the role of NR2F2 and Prox1 in maintaining LEC phenotypes.

Activated Notch-induced transcriptional profile modulation in human primary dermal lymphatic endothelial cells

(Submitter supplied) Human Notch1 intracellular domain (NICD) was overexpressed in human primary lymphatic endothelial cells (LECs) for 10 and 24 hours by adenovirus. Unique functions for Notch4 in murine embryonic lymphangiogenesis

(Submitter supplied) To assess DLL4/Notch LEC signaling, we seeded HdLECs on DLL4FC-coated or control FC-coated plates and collected RNA after 6 hours which was subjected to mRNA sequencing. Relative to the HdLECs seeded on FC-coated plates, DLL4FC significantly altered the expression of 675 genes with a padj <0.05. 69 genes were induced 1.2-fold while 68 genes were suppressed 1.2-fold. Analysis of the top 30 upregulated genes revealed DLL4/Notch signaling upregulated the expression of known direct effectors of Notch signaling, Hey1, EphrinB2, Hes4, Dll4, and Hes1, as well as key lymphangiogenic genes, Gja4 (Cx37), Cxcr4, Gja1 (Cx43), Ccl2, Ackr3, and Sema3g). more...

(Submitter supplied) 48hpf kdrl:egfp zebrafish brains were dissociated using Liberase and pdgfr beta egfp postive cells FAC sorted into Trizol LS. RNA was extracted and amplified before sequencing. Samples were prepared in triplicate, with 6 brains used per samples.
