Single cell RNA sequencing of bovine blastocysts: Early cellular differentiation

(Submitter supplied) Differentiation of totipotent embryonic cells into the first cell lineages (trophectoderm (TE) and inner cell mass (ICM), and further into epiblast (EPI) and primitive endoderm (PE)), although well studied, is only beginning to be understood at the molecular level. An in depth understanding of the regulation of these early differentiation events would enable improvements in the quality of in vitro produced embryos, as well as the study and application of bovine embryonic stem cells. more...

PGE2 supplementation of oocyte culture media improves the developmental and cryotolerance performance of bovine blastocysts derived from a serum-free in vitro production system, mirroring the inner cell mass transcriptome

(Submitter supplied) We focused our investigation of an embryo's response to different in vitro environments on changes to the transcriptomic profile of the inner cell mass (ICM), i.e. that part of the blastocyst giving rise to the epiblast and then to all future intraembryonic tissues. We used RNA-sequencing analysis to examine the gene expression of ICM derived under the different IVP conditions. The study evaluated the effects of a serum-free (SF) IVP system including defined oocyte culture media supplemented or not with PGE2, on gene expression of the ICM when compared with standard serum-containing (SC) IVP.

BBr002: A genomic landscape of enhanced bovine pluripotency

(Submitter supplied) Genes and signaling pathways involved in pluripotency have been studied extensively in mouse and human pre-implantation embryos and embryonic stem (ES) cells. The unsuccessful attempts to generate ES cell lines from other species including cattle suggests that other genes and pathways are involved in maintaining pluripotency in these species. To investigate which genes are involved in bovine pluripotency, expression profiles were generated from morula, blastocyst, trophectoderm and inner cell mass (ICM) samples using microarray analysis. more...

Lineage regulators TFAP2C and NR5A2 function as bipotency activators in totipotent embryos

(Submitter supplied) During the first lineage segregation, a mammalian totipotent embryo differentiates into inner cell mass (ICM) and trophectoderm (TE). However, how transcription factors (TFs) regulate this earliest cell fate decision in vivo remains elusive, with their regulomes primarily inferred from cultured cells. Here, we investigated the TF regulomes during the first mouse lineage specification across 6 stages, spanning the pre-initiation, initiation, commitment, and maintenance phases. more...

Functional role of GATA3 and CDX2 in lineage specification during bovine early embryonic development

(Submitter supplied) Current understandings of the initiation of the trophectoderm (TE) program in mammalian embryonic development lacks evidence of how TE-associated factors such as CDX2 and GATA3 participate in bovine lineage specification. In this study, we describe the effects of TE-associated factors on lineage specification marker genes such as SOX2, OCT4, NANOG, GATA6 and SOX17, assisted by a cytosine base editor system. more...

Gene expression analysis of in vivo originated bovine embryos fllushed out at 2- or 8-cell stages and cultured in vitro till blastocyst stage

(Submitter supplied) There is an inevitable need for combining in vitro and in vivo culture systems during specific developmental time points to facilitate a comprehensive understanding of early embryo development which would yield insights into the molecular pathways controlling early development and to improve our knowledge in regulation of embryonic development. In current study, we aimed to understand the influences of in vitro culture conditions on bovine blastocyst originated in vivo at 2- or 8-cell stagre. more...

Lipid stores and lipid metabolism associated gene expression in porcine and bovine parthenogenetic embryos revealed by immunostaining and RNA-seq [Ss]

(Submitter supplied) Compared to other mammalian species, porcine oocytes and embryos are characterized by large amounts of lipids stored mainly in the form of droplets in the cytoplasm. The amount and the morphology of LD change throughout the preimplantation development however, relatively little is known about expression of genes involved in lipid metabolism of early embryos. We compared porcine and bovine blastocyst stage embryos as well as dissected inner cell mass (ICM) and trophoblast (TE) cell populations with regard to lipid droplet storage and expression of genes functionally annotated to selected lipid Gene Ontology terms using RNA-seq. more...
