Liposarcoma response to doxorubicin in vitro

Analysis of various types of liposarcomas treated with doxorubicin in vitro. While doxorubicin is an established chemotherapeutic for treating liposarcomas, the response to this drug is low. Results provide insight into the molecular basis of the resistance of liposarcomas to doxorubicin.

Heterogeneous in vitro effects of doxorubicin on gene expression in primary human liposarcoma cultures

(Submitter supplied) Doxorubicin is considered one of the most potent established chemotherapeutics in the treatment of liposarcoma; however, the response rates usually below 30%, are still disappointing. This study was performed to identify gene expression changes in liposarcoma after doxorubicin treatment. Cells of 19 primary human liposarcoma were harvested intraoperatively and brought into cell culture. Cells were incubated with doxorubicin for 24 h, RNA was isolated and differential gene expression was analysed by the microarray technique. more...

Genomic characterization of well differentiated/dedifferentiated retroperitoneal liposarcoma

(Submitter supplied) An integrated profiling approach was performed to define molecular alterations associated to the aggressive behavior of retroperitoneal dedifferentiated liposarcoma. In particular, matched well and dedifferentiated components as well as normal adipose tissues, obtained from the same tumor sites, were comparatively investigated to higlight differences in gene expression.

MicroRNA expression profiles distinguish liposarcoma subtypes and implicate miR-145 and miR-451 as tumor suppressors (validation)

(Submitter supplied) Liposarcomas are rare, heterogeneous and malignant tumors that can be divided into five histological subtypes with different characteristics and clinical behavior. Treatment consists of surgery in combination with systemic chemotherapy, but nevertheless mortality rates are high. More insight into the biology of liposarcoma tumorigenesis is needed to devise novel therapeutic approaches. MicroRNAs (miRNAs) have been associated with carcinogenesis in many tumors and may function as tumor suppressor or oncogene. more...

Whole-transcript expression data for liposarcoma

(Submitter supplied) Liposarcoma is the most common soft tissue sarcoma, accounting for about 20% of cases. Liposarcoma is classified into 5 histologic subtypes that fall into 3 biological groups characterized by specific genetic alterations. To identify genes that contribute to liposarcomagenesis and to better predict outcome for patients with the disease, we undertook expression profiling of liposarcoma. U133A expression profiling was performed on 140 primary liposarcoma samples, which were randomly split into training set (n=95) and test set (n=45). more...

Cell-type specific responses to chemotherapeutics in breast cancer

(Submitter supplied) Recent studies have identified clinical subtypes of breast tumors that arise from different cell types and have different outcomes in response to chemotherapy. Tumors derived from basal epithelium have a poorer prognosis than tumors derived from luminal epithelium. To gain insight into differences underlying this disparity, we treated cell lines derived from basal epithelium (immortalized human mammary epithelial cells) and those derived from luminal epithelium (MCF-7 and ZR-75-1) with two chemotherapeutics commonly used in the treatment of breast cancer. more...

Doxorubicin resistance in a novel in vitro model of human pleomorphic liposarcoma associated with ALT

(Submitter supplied) Soft tissue sarcomas are a diverse set of fatal human tumors where few agents have demonstrable clinical efficacy, with the standard therapeutic combination of doxorubicin and ifosfamide showing only a 25-30% response rate in large multi-institutional trials. Although liposarcomas are the most common histological form of adult soft tissue sarcomas, research in this area is severely hampered by the lack of experimentally tractable in vitro model systems. more...
